Glycine Dosage Guidelines and Considerations

Glycine is a versatile, non-essential amino acid that acts as a major inhibitory neurotransmitter and a vital structural component for connective tissues. In clinical practice, its application is dose-dependent based on the targeted physiological system.

Target Concern Recommended Dosage Clinical Rationale
Sleep Quality 3g (30–60 mins before bed) Facilitates sleep onset by lowering core body temperature and modulating NMDA receptors.
Metabolic Health 3g – 5g daily Supports insulin sensitivity and helps dampen postprandial glucose spikes.
Connective Tissue 5g – 15g daily Provides the primary substrate (1/3 of total amino acids) for endogenous collagen synthesis.
Neurological Support 0.8g/kg body weight High-dose therapeutic intervention used in specific clinical research for refractory cases.

Clinical Considerations:

  • Administration: For doses above 3g, a powdered form is preferred to reduce pill burden and improve patient compliance.

  • Gastrointestinal Tolerance: While generally safe, doses exceeding 15g may cause mild osmotic diarrhea or nausea in sensitive patients.

Glycine and N-Acetyl Cysteine (GlyNAC) Synergy

The combination of Glycine and N-Acetyl Cysteine (NAC), known as GlyNAC, is a potent clinical strategy for restoring intracellular glutathione—the body’s master antioxidant.

The Glutathione Triad: Glutathione is synthesised from three amino acids: Cysteine, Glycine, and Glutamate.

  1. Cysteine (via NAC): Often the rate-limiting precursor for synthesis.
  2. Glycine: Frequently becomes deficient in ageing or chronic inflammatory states, creating a “secondary” bottleneck for antioxidant production.

Therapeutic Benefits of GlyNAC:

  • Mitochondrial Function: Improves fatty-acid oxidation and ATP production.
  • Oxidative Stress: Reduces biomarkers of cellular damage, particularly in “inflammation” protocols.

  • Metabolic Restoration: Research indicates improvements in muscle insulin sensitivity and systemic inflammation.

Dosing for GlyNAC:

  • Standard Protocol: A 1:1 ratio of Glycine to NAC.

  • Dosage Range: 1,200mg to 5,000mg of each, typically administered in divided doses with meals to enhance NAC tolerability.

Glycine does not act in isolation. Its clinical efficacy is often modified by its relationship with other amino acids:

  • L-Glutamine: Both are involved in maintaining the integrity of the intestinal mucosal barrier. Combined supplementation is often used in gut repair protocols to support epithelial cell turnover.
  • L-Serine: Serine is a precursor to Glycine. In some neurological contexts, Serine may be used as a metabolic alternative, though Glycine remains the direct neurotransmitter of choice for inhibitory signaling.
  • L-Methionine: Glycine acts as a buffer for excess methyl groups. High intake of methionine (prevalent in meat-heavy diets) can increase the demand for glycine to manage homocysteine clearance.

Practitioner Safety & Contraindications

  • Cystinuria: NAC-containing protocols are contraindicated in patients with this rare genetic condition due to the risk of cystine stones.
  • Peptic Ulcers: Use caution with NAC in patients with active ulcers as it may reduce the viscosity of protective gastric mucus.
  • Medication Interactions:
    • Clozapine: High-dose Glycine may interfere with the efficacy of this antipsychotic medication.
    • Nitroglycerin: NAC can potentiate the vasodilatory effects of nitroglycerin, potentially leading to severe hypotension or headaches.
  • Authorisation Note: As a practitioner-only restricted product, ensure dispensing follows HealthMasters protocols for quantity and duration limits.
